Toluidine Blue is a normal thiazine metachromatic dye with very high similitude to acidic components and thereby affecting/staining the tissues which are rich in DNA and RNA. Because of this feature it have a wide number of applications both in living tissues and a different and special stain owing to its metachromatic property. In vivo, toluidine Blue has been used to identify dysplasia and carcinoma produced in the oral cavity. The components such as mast cells granules, cartilage and mucins are treated with toluidine blue with a aim of highlighting them. Toluidine is known by all because of its various medical applications. It was discovered by William Henry Perkin in 1856. After its discovery it is has been popularly used in the dye industry. Toluidine Blue is also known as methylaniline or aminotoluene. ortho-toluidine, para-toluidine, and meta-toluidine are basically the three isoforms of toluidine blue. It is majorly used in mucosal lesions as a vital stain. Also it is majorly used in as applications in the tissue section to highlight the stain according to its metachromatic property.

The two techniques involved in the vital staining are the intravital staining and the supravital staining. Toluidine Blue is very effective in detecting relative differences between normal and malignant cells and tissues . Also it is very useful in highlighting the vital stain of malignant oral lesions. Also it is very helpful in identification of early lesions which are being missed in normal clinical examinations. Most of the dyes stains on the tissue with the same colour with different degrees of intensity. The reaction of the staining is is called as metachromasy and the tissue showcase metachromasia. There are other tissues components such as mucin, cartilage and mast cells granules which have exhibited the metachromasia. Toluidine blue is one dye which is being used in the tissues section for the identification of the various cells.
